To take advantage of the size of the poster and its ability to draw viewers is based on the size of the poster, you should concentrate on using large images and eye catching colors. It is wasteful to have a huge canvas like a poster and then crowd tons of small text and imagery on to it. No one is going to read a 2 foot by 3 foot poster filled with tiny type. But they will more than likely read the type on a poster which has already drawn their attention with a large image.
